Bigger the better for performance, but that cuts into material thickness of the panels and strength of the joints.

Routing the edges will help prevent the sharp corner from fracturing if there is an accident. The rounded edge also helps with refraction of the drivers on the sharp corners. Grab a scrap piece and do one side with the quarter inch round over, the other with the half inch round over and see which one you like the look of better.

I believe the 1/2” will give better performance and if you are veneering the front baffle and sides as one piece, it will be easer to make the 1/2” over the 1/4” in my opinion.

Mike, what's the largest round over you can go before it weakens the corners too much? Assuming the standard 3/4" mdf.

Danny's recommendation is 3/8" which is right in the middle of both 1/4" and 1/2"

But 1/2" will be fine. so long as you're using 3/4" MDF for your panels.

you can mitigate any weakness from 1/2" by adding small wedges to the corners, or adding additional glue along the inside edge (so long as the wedges don't interfere with the woofer cut out, which also needs 3/8" round-over on the inside edge)
