As far as H-Frame designs go

All things being equal, will these measure the same? Will it be audible? The walls are transparent to show the differences only.

I am the wrong cat to comment on the science and the measuring, but there are others who know more that will share I hope, I think it is a cool question.

For me, the 2nd version is vibration waiting to happen.  I like to see short distances between the braces on OB designs, vibration in bass can be a frustration.

Good point. Please assume both will be properly braced for vibration reduction. The focus is on the baffle division.

what is the purpose of the drivers facing in opposite directions and would they need to be wired out of phase  to reduce cancellations.

If the response of all drivers would be identical, taking away the divisions would make no difference at all. Now that you have two drivers reversed, there are slight differences in response which will lead to (very?) small adjustments.
Certainly you don't win or loose any dB with or without the dividers. But the vibration argument is a very strong one!

I believe that removing the dividers will change the loading on the drivers, so the tuning of the enclosure will change.  You would need to model it w/ something like Martin King's MathCAD worksheets to find out how much the effect would be.

As far as dipole cancellation goes, the two configurations will be the same since the front/back distance is unaltered.

In the upper example you have created cavities that will yield small resonant peaks....but probably at a frequency above your intended usage range so it should not be a problem.

The thing I'd be concerned with in either design is vibration generation.  It may not be a problem if your listening room floor is solid, but I've seen (heard) "H"-baffle configurations cause issues on more flimsy floors.
