I'm test driving my new GIK 244 panels and I'm using them at the first reflection point.  Is there any advantage to using the 242's for this over the 244's?

Not at all.  In reality, depending on the boundary interactions happening in your room, the 244's may well be superior.

How could I tell for sure which would be better between the two?  Also I have them sitting on the floor and they are approx the same height as my speakers.  Do the panels perform better if they are off the ground a few inches or on the floor?

Use the mirror trick and place the center of the panel in that location.  You are splitting hairs with which of the two to use on the side walls.  I'd put the thicker panels near/behind the speakers for a little more bass absorption and the thinner on the sides.  But until you get real serious about 8 inch or better bass trapping it will be unnoticeable.
